The ER-localized version of FMNH2, MAM20019r, participates in 5 reactions:
MAR20021
MAR20023
MAR20025
MAR20027
MAR20029
In each case, FMNH2 acts as a reactant in a non-reversible reaction. As a result, there is no synthesis route for the metabolites, and all of these reactions must have a flux of 0 under steady-state. I don't know whether this should be addressed by the reactions having reversibility, or whether FMNH2 transport into the ER is missing, or what.
The FMNH2 in these CYP2E1 reactions comes from the Rhea reactions' "reduced [NADPH–hemoprotein reductase]", taken as free FMNH2. The reductase takes its electrons from NADPH, as in the model's other ER CYP reactions and in CYP2E1's MAR20031.
Path forward:
MAR20021/23/25/27/29 (ER): use NADPH + H+ → NADP+ instead of FMNH2 → FMN. The reactions stay blocked, because their hydroxylated products have no known fate: no export or further metabolism is established, so no transport or exchange reactions are added just to unblock them.
MAR20022/24/26/28/30 and MAR20032 (i): deprecate. They are copies in the intermembrane-space compartment, where none of their metabolites takes part in any other reaction.
Follow-up to #1094, in #1105 (open): The CYP2E1 (omega-1)-hydroxylations MAR20021-MAR20029 get names and the references UniProt uses (PMID:18577768, PMID:10553002); MAR08034 gets CYP3A4 next to CYP2E1 (PMID:9348445). Details and evidence are in the comment on #1094.
